Polymorphic DNA loci were analyzed by polymerase chain reaction of DNA synthesis, followed by electrophoretic detection. Polymorphism in genes involved in ethanol metabolism was studied/RESULTS: The patents with ACP had mutations in the genes involved in ethanol metabolism. The study results of the rate of the alleles and genotypes of ADH, ALDH, and CYP2E1 genes were controversial. However, investigating the combinations of the genotypes of ADH and ALDH revealed that the patients having ADH1B*47His and ALDH2*2 genotypes had a much higher risk for ACP and constituted more than 50% of the ACP patients. With this combination, ACP took its course with more evident structural and functional disorders of the pancreas. The CYP2E1 С/С homozygotes had an especially high risk of ACP/CONCLUSION: The investigation perspectives are to increase the number of examined patients and apparently healthy individuals and to find correlations between clinical, laboratory, and instrumental parameters and genotyping findings. The results of these investigations can define the possible strategy of prevention of CAP.</p></abstract><trans-abstract xml:lang="ru"><p>Резюме. Цель исследования. Изучить полиморфизм генов алкогольдегидрогеназы (ADH), альдегиддегидрогеназы (ALDH), цитохрома CYP2E1 у больных хроническим алкогольным панкреатитом (ХАП) и провести сопоставление с лабораторно-инструментальными данными. Материалы и методы. Обследовали 72 больных ХАП и 80 здоровых лиц. Анализ полиморфных локусов ДНК осуществляли методом полимеразной цепной реакции синтеза ДНК с последующей электрофоретической детекцией. Изучали полиморфизм генов, участвующих в метаболизме этанола. Результаты. У больных ХАП имеются мутации генов, участвующих в метаболизме этанола. Результаты изучения частоты аллелей и генотипов генов ADH, ALDH и CYP2E1 противоречивы. Однако при изучении сочетаний генотипов ADH и ALDH выявлено, что у больных с генотипом ADH1B*47His и ALDH2*2 значительно повышен риск развития ХАП, и они составляют более 50% больных с алкогольным поражением поджелудочной железы. При наличии такого сочетания ХАП протекает с более выраженными структурными и функциональными нарушениями поджелудочной железы. Гомозиготы CYP2E1 С/С имели особенно высокий риск формирования ХАП. Заключение. Перспективы исследования состоят в увеличении числа обследованных больных и практически здоровых лиц и в поиске корреляций между клиническими, лабораторными, инструментальными показателями и результатами генотипирования.
